Transgenic Core Facility (TCF)
Focus
The Transgenic Core Facility (TCF) provides a centralized resource for the generation of mouse models to understand human diseases and assist in developing novel therapies. We provide full support from transgenic design to the generation of the mouse models, depending on the need of the researchers. We make use of the full toolbox of CRISPR/Cas9 applications to generate complex genome engineered mice. Furthermore, the TCF provides standard services such as rederivation, sperm testing and conservation of mouse lines.

Services
Services for standard mouse strain maintenance
- Rederivation of mouse strains from cryoconserved sperm and embryos, incl. IVF
- Sperm and embryo cryoconservation, optionally with rederivation test
- Embryo transfer into foster animals for strain cleanup
Established transgenic services
- Design and consulting on transgenes and strategies
- Generation of complex targeting constructs with next generation conditional knock-out strategies
- Assistance for license application from the authorities for novel mouse lines
- Microinjection of nucleic acids and riboprotein CAS9 complexes into zygotes (pronucleus or cytoplasm)
- Ex vivo zygote electroporation with riboprotein CAS9 complexes
- ES cells gene targeting with screening for clonal selection
- ES cell aggregation with diploid/tetraploid host cells for embryo and strain generation
- ES cell microinjection of blastocysts
